Neighborhood Spatial Analysis was performed using a cell spatial analysis pipeline which determined the cellular composition within a 30-μm radius around individual cells and performed unsupervised k-means clustering (Python Scikit-learn, version 0.21.2) followed by manual annotation using the main cell population in each cluster. Fractal Dimension (FD) of each marker was analyzed using the box count method implemented in FIJI Fraclac Plug. Each biomarker’s IMC image was binarized first and the FD was analyzed. The results were summarized using the average FD for each cohort. To compare the FD between the two cohorts in this study, each Biomarker’s FD was normalized based on its tumor cell biomarker (PanCK).
